The Orica-GreenEDGE rider won the final stage and used time bonuses to make up a three-second deficit on Italian Monia Baccaille to take the overall title by just one second.
Hoskins had also won the opening stage two days ago.
The Orica-GreenEDGE team including Olympic road cycling hopefuls Alexis Rhodes, Amanda Spratt and Shara Gillow finished second in the overall classification.
Hoskins will ride on the track at the Olympics as a member of Australia's team pursuit squad, which has made substantial improvement this year to put pressure on Great Britain.
Australia won silver behind the British in the team pursuit at last year's world championships in Melbourne.
On the road, Hoskins also won the Jayco Bay Classic overall title in January.
