Manzano was in second place behind five time titleholder Joanne Carter after the short program, registering a total segment score of 45.50 points to the Sydney skater's 54.36.
But in the free skating, she was awarded 85.85 points by the judges for an overall score of 131.35 points.
Carter's free program 75.70 points gave the silver medal on an overall score of 130.06 points, with South Australian junior Janna Kupec in the bronze medal position with 81.82 points.
In the men's event, Sean Carlow captured his first national title with 107.70 points from defending champion Brad Santer on 103.38 points.
The Pairs title went to Emma Brien and Stuart Beckingham, while Natalie Buck and Trent Nelson-Bond won their fifth straight national Ice Dancing championship.
